Immobilization of a his(6x)-protein

Hello everyone!

I am fairly new to SPR and have a question about the immobilization of a purified His(6x)-protein. The size of the protein is about 35 kDa.

In the literature I found immobilization protocols using a NTA-Chip. Can I also use a CM5 oder HC200M-Chip and a covalent immobilization using EDC/NHS? Or isn't it possible because of the His-Tag?

Hi Jana,

You can use the amine coupling on alomost all proteins provided that the protein has free lysines and that they are not only on the interaction site. Having a 6xHIS-tag will not impair amine coupling.
