C1 - Flat carboxylation

Sensor chip C1 has a flat carboxymethylated surface. Provides the same functionality as Sensor chip CM5 but has no dextran matrix (the carboxyl groups are attached directly to the surface layer). The absence of a surface matrix makes Sensor chip C1 less hydrophilic than Sensor chip CM5 that, for some samples, may increase non-specific binding. This may be diminished by adding 0.2 mg/ml BSA to the running buffer. The flat carboxylated surface may be of value in work with large analytes such as cells and virus particles, and can be used to test the influence of the CM5 dextran matrix on kinetic determinations. Experimental protocols follow the same principles as applied to Sensor chip CM5. The absence of a surface matrix will result in an immobilization yield that is approximately 10% of that obtained on Sensor chip CM5 under comparable conditions.

To clean the surface before immobilization two injections of 1-2 minutes with freshly prepared 0.1 M Glycine-NaOH pH 12 with 0.3 % Triton X-100 are done. Add an EXTRACLEAN command after the injection commands.
