Regeneration of Au chip was created by hnnigel
Hi,

I am currently using BiaCore to see whether growth factors can be bound to the protein immobilised Au chip.
Our group used to use 1 or 2 M NaCl buffer (inject short time at high flowrate) to regenerate the used Au chip (it has protein, BSA/PBS and GFs), and it used to bring down the RU to the initial RU value . However, 1 or 2 M NaCl buffer cannot regenerate the chips anymore.

I would like to know whether there are another buffers to regenerate Au chips.

Thanks,
Ha Na

I don't know special regeneration solutions for AU surfaces.
Regeneration should be effective and not damaging your ligand. What we do is try first the mildest solution and check if binding still occurs. Then go harsher for better regeneration.
Look at www.sprpages.nl/kinetics/regeneration and the cocktail regeneration.
Start with 6x dilutes Ionic solution, then 3x. Then combine with lower or higher pH etc.

Kind regards
Arnoud
