Unusual sensorgram shape

The problem with the immobilizing the anti-His antibody and capturing the ligand is that you don't know how well the surface is blocked. Even when you make a reference and block this surface, it seems to me that you introduce a lot of uncertainty in the surface.
I would recommend immobilizing the ligand directly on the surface even when this means that it is randomly orientated.
