BSA in running buffer

BSA in running buffer was created by ebelcheva
Hi,
The running buffer used in my standard conditions contains .5 mg/mL BSA. I noticed in few publications, some add the BSA following the filter and degassing step. I am wondering if anyone has experience with adding BSA to their running buffer to address non-specific binding and the potential implications of adding the BSA prior to degassing

Thank you
Ellie

Hi Ellie,
Degassing a solution with high protein can give some foam on top of the solution.
I make normally a stock solution of (standard) running buffer and degas an aliquot just before use. After degassing I add some concentrated BSA solution (filtered) to get my desired BSA concentration and mix gently. The same applies for adding detergents.
Kind regards

Arnoud
