hi,all
I am a new user about SPR. Recently, our laboratory conducted an experiment, but I am not sure whether the result is reliable. I would like to ask for your advice.
So here's my experimental condition :
CM5-coupled anti human IgG Fc (3&4 channel)
4 channels capture hFc-labeled proteins with a molecular weight of 40.7KD.Then 3&4 channels flow through the analyte protein, labeled with his, with a molecular weight of 38.1KD.
RL=500RU.
I think the amount of capture is too high for the protein, and the binding signal of the analyte may be non-specific binding signal.I tried to captue ligand at 70RU, but the analyte did not bind the signal at 1000nM.

Difficult to say if this is proper interaction since the bulk-effects are fairly high. It is better to get a better match between flow buffer and analyte injection.
I would not try to kinetically fit these data because of the large buffer jumps. The lack of curvature in the curves result in very unreliable values.
