Hi,

We are trying to do SPR for irreversible covalent small molecule ligands (~50 different ligands) so need a regenerate-able SPR system. We have recently tried Biacore's CAP system, but despite good binding of the ligand, we were unable to detect analyte binding, even for our positive controls.

As a result we would like to try a thiol coupling system, with the advantage that this may give us some higher ligand response and therefore Rmax as well. Hopefully this will allow us to detect analyte binding.

As such, I was just wondering if anyone has had any experience with frequently-regenerated thiol coupling and could offer some advice on optimal conditions in addition to the SPR pages section on this.
