Hi Arnoud,
I am using Biacore T200 and currently am working with a very high affinity molecule, I am using CM5 chip. I am facing severe problem with the dissociation as it is going out of my instrument specifications. I tried all the possible ways which includes single cycle kinetics, MCK, varied dissociation time, 3 hrs of dissociation, Changing the buffer constituents, Buffer pH but nothing has worked so far. My QC tabs are failing and if my QC tabs passes my results are not consistent. Please do let me know a solution for this.

Hi,
Ik think you have to switch to some different techniques. You can look up
BLI for kinetics or MST or ITC for determination of the KD alone. Look at www.biapages.nl for a short introduction of these techniques.
