Opposite Concentration Order on Sensorgram

Hello,

I have a query about results from the attached sensorgram. The concentration range was initially rising, then as it got to 1.25uM concentration, it began to decrease in RU instead of increasing. Can you please help me as to why this might happen?

The experiment was carried out on an NTA chip on a Biacore T200. I used 150mM NaCl, 20mM HEPES, 0.01% P20 and 50uM EDTA in my buffer and used 0.5uM Nickel sulphate and EDC/NHS when immobilising. The analyte is a LMW compound, and association time was 60s and dissociation time was 120s. I used 10 start-up cycles of blank buffer too.
