anyone, we think some non-specific binding is occuring and what to try out adding more P20 surfactant to the existing HBS-P buffer but not sure how much to add. any suggestions? currently the HBS-P has 0.5% v/v of P20 in it. Also would adding BSA to samples help? Any suggestions would be most helpful.....Thanks.

0.5% is alrealdy quite a lot. You could try adding BSA to the running buffer (say 0.5 mg/ml for a start) and some extra salt (0.5M) or dextran. Working at low concentrations can also help.
