looks like my initial post did not go through....

Anyway, has anyone done any work on Peptides? The mol. wt. of my compound is around 1500-2000. Looks like I have adequate binding response on the Ab up front but the Ag does not seem to bind, relatively flat (conc. is ~200nM for the top end of the 5 concentration prepared). Any suggestions?

Hi mtarca,

Is the Ag hydrophobic or ampipathic and does it aggregate? Hydrophobic molecules are likely to aggregate in the hydrophilic running buffer, and this will prevent binding to ligand. This of course assumes that the analyte in question definately does bind the ligand.
